Yeast Two-Hybrid (Y2H) Screening Service

The United States Department of Agriculture (USDA) Agricultural Research Service (ARS) requires a Yeast Two-Hybrid (Y2H) Screening Service to conduct research on the feasibility of new virus transmission management approaches. The contractor will design, construct, and execute a yeast two-hybrid (Y2H) screening pipeline for systematic identification and validation of virus–aphid protein–protein interactions. The scope of work includes designing and constructing viral bait constructs, generating high-quality aphid cDNA prey libraries, conducting large-scale Y2H screening, and providing technical reports and data deliverables. * The contractor will prioritize viral proteins and/or domains for use as Y2H baits in consultation with government scientists. * The contractor will design and construct three viral bait clones suitable for yeast two-hybrid screening. * The contractor will generate high-quality aphid cDNA prey libraries optimized for detection of virus–vector protein–protein interactions. * The contractor will conduct large-scale Y2H screening to identify aphid proteins interacting with viral bait proteins.
